抗CD4单抗-PAP免疫毒素对T淋巴母细胞样CEM细胞的体外效应

摘要 【目的】构建抗CD4单克隆抗体-商陆抗病毒蛋白(PAP)免疫毒素(抗CD4-PAP),分别在体外测定其对T淋巴母细胞样细胞CEM和对照Raji细胞的细胞毒杀伤效应。【方法】以SPDP为交联剂,将纯化的抗CD4单抗与商陆抗病毒蛋白交联成抗CD4-PAP免疫毒素,在体外分别与CEM和Raji细胞进行杀伤试验。【结果】10-9mol/L抗CD4-PAP对CEM细胞产生的细胞毒效应约为60%,而对Raji细胞的细胞毒效应低于10%。抗CD4-PAP免疫毒素在体外对骨髓单个核细胞的CFU无抑制效应,【结论】抗CD4-PAP免疫毒素对T淋巴母细胞样CEM有明显的抑制效应,可能替代抗CD3单抗作为器官移植抗排斥反应及导向免疫治疗T淋巴细胞白血病的临床治疗研究。 To construct immunotoxin by conjugating the monoclonal anti-CD4antibody to pokeweed antiviral protein(PAP),and to examine its cytotoxicity eff ect on T lymphoblastic cell(CEM)and Raji cell respectively.Purified anti-CD4monoclonal antibody was linked with PAP using SP DP to construct anti-CD4-PAP immunotoxin.The cytotoxicity of immunotoxin to CEM and Raji cells was carried on.Anti-CD4-PAP immunotoxin showed stronger killing effect on CEM than o n Raji cells,and had no inhibition effect on CFU of bone marrow cells.[Conclusion]Anti-CD4-PAP immunotoxin showed di stinct inhibition effect on CEM cells.It could be used as a substitute for anti-CD3monoclonal antibody for T cell l eukemia and organ transplantation.
