摘要
目的 :观察Li-Pilocarpine大鼠癫痫模型在慢性期出现反复自发性癫痫发作过程中 ,大鼠海马在1H -MRS上的进行性变化 ,从而探索癫痫产生的病理生理机制。材料和方法 :分别在成功诱发癫痫持续状态后大鼠的3h至8周的不同时间点 ,提取海马内组织标本 ,行 1H -MRS检查 ,测量海马内N -乙酰天门冬氨酸 (NAA)、胆碱(Cho)、肌酸 (Cr)、谷氨酸 (Glu)和γ -氨基丁酸 (GABA)水平 ,并与正常对照组相比较 ,观察上述指标在急性期至慢性期的动态变化。结果 :在急性期 ,谷氨酸水平的升高 ,此后海马内NAA/Cr水平开始降低 ,3d后海马内NAA/Cr水平明显降低。海马内Cho/Cr在慢性期明显升高 ,此时海马内GABA/Cr水平较正常对照组水平明显减低。结论 :GABA能神经元的丢失 ,与慢性期反复自发性癫痫发作的出现有关。
Purpose:This study investigates the progressive changes before appearing spontaneous recurrent seizures at the chronic period in the epilepsy model by 1 H-MRS of hippocampus and search for basic mechanisms of epileptogenesis.Materials and Methods:We monitored all the rats from3hours to8weeks after the onset of status epilepticus.The hippocampus extract was measured using 1 H-MRS to define the levels of NAA,Cho,Cr,Glu and GABA in it,which compared the with normal control groups to detect the changes from the acute to chronic period.Results:At acute period,there was a highly elevaˉtion of Glu/Cr level,then the NAA/Cr level in the hippocampus began to decrease.The NAA/Cr level decreased obviously3days after the status epilepticus.The Cho/Cr level in the hippocampus was elevated at chronic period,and the GABA/Cr level in the hippocampus decreased obviously compared to normal controls.Conclusion:The loss of GABAergic neurons is related to the onset of spontaneous seizures at the chronic period.
