高效液相色谱法测定脂质体中硝酸益康唑的含量及其理化性质的考察

Determination of the drug content in econozole nitrate liposome by HPLC and study on its physical proprieties

目的 :高效液相色谱法测定硝酸益康唑脂质体中药物的含量并考察其理化性质。方法 :建立了用色谱预处理柱除磷脂测定脂质体中药物含量的HPLC法 ,并用此法测定了硝酸益康唑的溶解度和分配系数。结果 :此法可以除去大部分磷脂 ,具有较好的专属性、回收率和精密度 ;硝酸益康唑在水 ,生理盐水 ,2 0 %PEG生理盐水中的溶解度分别为 (879.4± 4 2 .9) ,(36 1 .6± 2 3.8) ,(30 0 6 .5± 6 0 .4 )mg·L-1 ,油 /水 ,表皮 /水和真皮 /水分配系数为 4 5 .4 ,4 .82× 1 0 5和 5 .31× 1 0 5。结论 :此法适于测定硝酸益康唑脂质体中药物含量 ;硝酸益康唑对表皮和真皮有较强的亲和力 ,适于皮肤给药。

OBJECTIVE To determine the drug content in econozole nitrate liposome by HPLC and study the physical proprieties.METHODS The HPLC determination of the drug content in liposome was developed the drug solubility and partition coefficients were determined.RESULTS The PT-C 18 can remove most phospholipid, the specificity、recovery、precision of the method were good, The solubility of Econozole nitrate in water、saline water、20%PEG saline water were ( 879.4± 42.9), ( 361.6± 23.8), ( 3 006.5± 60.4) mg·L -1 respectively and the oil/water、 epidermis/water、dermis/water partion coefficients were 45.4, 4.82×105 and 5.31×105 respectively.CONCLUSION The HPLC method is suitable for the determination of in econozole nitrate liposome. Econozole nitrate is applicable to skin because of its strong affinity.
