摘要
目的 :观察氨酚伪麻那敏 (GYT)片的解热、镇痛、抗炎、抗过敏作用和对血压、心率的影响等药效学 ,为评价其临床疗效和安全性提供依据。 方法 :以复方阿司匹林 (APC)片做对照 ,将GYT分为 2个剂量组 (10 0mg/kg ,5 0mg/kg)分别进行药效学动物试验观察。对试验数据进行统计学处理以评价其相应的药物效应 ,同时用简化机率单位法求其半数致死量 (LD50 )。 结果 :GYT和APC对家兔均有显著的解热作用 ,但二者的解热作用差异无统计学意义 (P >0 .0 5 ) ;GYT和APC的镇痛作用差异无统计学意义 (P >0 .0 5 ) ;2个剂量组的GYT均能使豚鼠抓鼻次数明显减少 ,其抑制率分别为 91.8%和 92 .5 % ,而APC(10 0mg /kg)的抑制率仅为 12 .5 % ;治疗量的GYT对家兔的血压和心率无明显影响 (P >0 .0 5 ) ;用简化机率单位法求得其半数致死量 (LD50 ) =1.31g/kg ,LD50 的95 %可信限为 (1.31± 0 .2 2 )g/kg。 结论 :GYT对组胺所致豚鼠鼻腔粘膜过敏反应的抑制作用及保护上呼吸道粘膜作用明显优于复方阿司匹林片 (P <0 .0 1)。
Objectives: In order to provide basis for clinic use and reliability evaluation, to study the pharmacodynamics of compound acetaminophen tablet (GYT) on anti-pyrogenic, anti-analgesia, anti-inflammation, blood pressure, cardiac rate and antianaphylaxis test. Methods: The pharmacodynamics test of two dose of GYT (100 mg/kg, 50 mg/kg) and control drug, aspirin phenacetin caffeine (APC), was done. LD 50 was calculated by using simplification probit. Results: There is no significant difference on anti-pyrogenic and anti-analgesia action between GYT and APC (P>0.05). The suppression rate on guinea pig anaphylaxis of GYT in two doses were 91.8% and 92.5% respectively. While the suppression rate on guinea pig anaphylaxis of APC (100 mg/kg) was 12.5% only. Under therapeutic dose, GYT didn't influence rabbit blood pressure and cardiac rate (P>0.05). The LD 50 of GYT was 1.31 g/kg with a 95% confidence limit of (1.31±0.22) g/kg. Conclusion: The suppression action on guinea pig anaphylaxis induced by histamin and protection on upper respiratory tract mucosa of GYT were higher than that of APC (P<0.01).
