摘要
目的 建立测定胡芦巴中 4 -羟基异亮氨酸的方法。方法 采用HPLC -ELSD法 ,IntersilODS - 3C18柱 ,水 -乙腈 -冰醋酸 (90∶10∶0 1)为流动相 ,柱温 4 0℃ ,流速为 1 0ml·min-1,ELSD气化室温度 110℃ ,气体流速 3 0L·min-1。结果 4 -羟基异亮氨酸在 0 0 6 85~ 0 2 392mg·ml-1范围内 ,线性关系良好 (r=0 9998) ,平均回收率 98 86 %。结论 该法简便、快速、准确性高、重复性好 ,适合于胡芦巴中 4
OBJECTIVE To establised a method for the determination of 4-hydroxyisoleucine contents in Trigonella foenungraecum. METHODS An HPLC-ELSD method was set up using Intersil ODS-3 C 18 column. The mobile phase was water-acetontrile-glacial acetic acid (90∶10∶0.1), with a flow rate of 1.0 ml·min -1 at 40℃.The temperature of drift tube heater was 110℃ and the gas was flowing at a rate of 3.0 L·min -1. RESULTS A good linearity was obtained in the range of 0.0685-0.2392 mg·ml -1 (r=0.9998) for 4-hydroxyisoleucine. The average recovery of 4-hydroxyisoleucine was 98.86%. CONCLUSION The method is simple and rapid with reproducibi^lity for the determination of 4-hydroxyisoleucine in Trigonella foenungraecum.
