摘要
目的 探讨采用DNA技术鉴定弱D15型。方法 根据文献和GenBank报道的基因序列 ,针对弱D15型基因 ,设计一对特异性引物 ,并引入一对内对照引物 ,建立简易的聚合酶链式反应 (PCR)技术检测弱D15型 ,之后用于检测 10份Rh阴性、10份Rh阳性、10份Del样品和 1例弱D15型DNA样品。结果 除 1例弱D15型样品检测为阳性外 ,其余均为阴性 ,显示PCR特异性检测弱D15型基因。结论 PCR方法简便、快速 ,能准确鉴定弱D15型 。
Objective To study on establishing a DNA-based method for the determination of weak D type 15.Methods One pair of primer was designed specific for weak D type 15 according to the literatures and sequences in GenBank. Subsequently a PCR method was established with a pair of internal control primer, and was evaluated with 10 Rh negative, 10 Rh positive, 10 Del samples and one weak D type 15 sample.Results The PCR results of all Rh positive, negative and Del samples were tested negative while the weak D type 15 sample was detected positive, which showed that the PCR was specific for the gene of weak D type 15.Conclusion The PCR is a simple and rapid method, which can determine weak type 15 correctly, and could be used for genotyping of weak D type 15 in routine clinical laboratories.
