摘要 目的 建立高效液相色谱荧光检测法测定脑脊液中丙泊酚 (异丙酚 )浓度的方法。方法 采用HypersilODS C18柱为分析柱 ;甲醇 水 β 环糊精 (6 8∶32∶0 .0 1)为流动相 ,流速为 1.0mL/min ;激发波长为 2 76nm ,发射波长为 310nm。 结果 异丙酚与脑脊液中其它成分分离良好 ,平均回收率为 10 4 .98% ,日内、日间RSD均小于 15 % ;脑脊液中异丙酚浓度在 10~ 32 0ng/mL范围内线性关系良好 ,r =0 .9992。结论 该法操作简便、结果可靠 。 OBJECTIVE To establish an assay method for propofol concentration in cerebrospinal fluid with high performance liquid chromatography.METHOD Hypersil ODS-C_(18) was packed in the analytical column; the mobile phase consisted of methanol-water-β-cyclodextrin (68∶32∶0.01); the flow rate was 1.0mL/min; excitation wavelength was 276nm, emission wavelength was 310nm.RESULTS Propofol was separated well from other components in cerebrospinal fluid. The mean recovery of propofol was 104.98% and the relative standard deviation (RSD) within a day and between days was all less than 15%. The assay linearity was determined over the range of 10 ~320 ng/mL in cerebrospinal fluid (r=0.9992).CONCLUSION The method was found to be simple and accurate, the experiment will supply reference methods for determination of propofol concentration in cerebrospinal fluid.
