中国汉族人群中TNF-α基因多态性与血脂关系的研究

Studies on association between TNF-α locus gene polymorphism and serum lipids in Chinese

目的 研究肿瘤坏死因子 -α(TNF -α)基因多态性与健康人群血脂谱改变的关系。方法 随机选择 182例湖北地区无血缘关系健康汉族人群 ,用聚合酶链反应限制性片段长度多态性 (PCR -RFLP)技术检测其TNF -α基因多态性 ,分析各基因型对血脂、脂蛋白、载脂蛋白的影响。结果 TNF -α - 2 38位点基因型在中国正常人群中的分布仅与日本人群中的分布较为接近 ,TNF -α - 86 3位点基因型的分布与日本人、高加索人中的分布差异无显著性。结论 在汉族人群中存在TNF-α基因多态性 ,且与血脂水平无相关性 (P >0 .0 5 )。

Objective: To examine the distribution of TNFα-238、-863 polymorphism in HuBei Han population.Methods; We use PCR-RFLP to examine the distribution of TNFαlocus gene polymorphism at-238、and -863 sites in HuBei Han population and detected the serum levels of TC、TG、apoA1、apoB、HDL-c and LDL-c to analyse the association between TNFα locus gene polymorphism and serum lipids in Chinese. Rusults:The TNFα-238 allelic and genotypic frequencies in Chienese were only the same as in Japnese.The TNFα-863 allelic and genotypic frequencies were the same as in Japnese and in Caucasian.The serum levels of lipids、lipoproteins and apolipoproteins have no difference in different genotypes.Conclusion:The TNFαpolymorphisms in Chinese were different from those observed in other ethnics.But we found no relations between the serum lipids and TNFα promoter gene polymorphism.
