摘要 目的 观察载阿仑膦酸钠骨水泥对骨水泥微粒诱导的骨吸收的抑制作用。方法 将制作的载阿仑膦酸钠骨水泥微粒和骨水泥微粒分别加入鼠骨髓单核细胞培养体系中 ,经 10 -8mol/L 1α ,2 5 (OH) 2 D3 和 10 -7mol/L地塞米松诱导培养 ,于培养 9、 12d时观察多核、抗酒石酸酸性磷酸酶 (tartrate resistantacidphosphatase,TRAP)染色阳性的破骨样细胞和形成的骨吸收陷窝数量。结果 鼠骨髓单核细胞培养体系中 ,加入载阿仑膦酸钠骨水泥微粒后 ,形成的破骨样细胞以及骨吸收陷窝数量明显少于不加或仅加入骨水泥微粒组 (P <0 0 1)。结论 载阿仑膦酸钠骨水泥能够释放出阿仑膦酸钠 ,抑制骨水泥微粒诱导的破骨样细胞的形成和骨吸收作用。 Objective To determine the inhibitory effect of Alendronate in bone cement on polymethylmethacrylate (PMMA) particles induced bone resorption. Methods Particles of PMMA mixed with or without Alendronate were generated and they were separately added to mouse bone marrow monocytes cultured with α-MEM containing 10 -8 mol/L 1α,25(OH)_2D_3 and 10 -7 mol/L dexamethasone. The extent of osteoclast-like cells differentiation was determined by measuring the number of tartrate-resistant acid phosphatase (TRAP) staining positive, multinuclear cells and the amount of lacunar resorption pits on bone slice.Results The addition of Alendronate-PMMA particles to mouse bone marrow monocytes cultures resulted in a marked decrease both in the number of TRAP positive, multinuclear cells and that of lacunar resorption pits compared with that no particles or PMMA particles only were added (P<0.01).Conclusion The bone cement carrying Alendronate can inhibit PMMA particles induced bone resorption.
