胎盘异铁蛋白对小鼠胚胎生长发育影响的研究

摘要 目的:探讨胎盘异铁蛋白对小鼠胚胎生长发育的影响。方法:将人早孕蜕膜细胞和小鼠2-细胞期胚胎置于含不同浓度胎盘异铁蛋白的培养液中共培养,倒置显微镜下观察记录胚胎数目并分级。结果:①发育至4-细胞期、8-细胞期及桑葚胚阶段鼠胚数的百分率比较,差异无显著性(P>0.05)。②在10 U/mL和100 U/mL浓度的胎盘异铁蛋白作用下,发育至囊胚期和孵出期鼠胚数的百分率明显提高(P<0.05)。③在1 000 U/mL浓度的胎盘异铁蛋白作用下,从2-细胞期发育至孵出期鼠胚数的百分率有所下降,部分细胞发生变性、退化和不规则分裂等现象,但无统计学意义(P>0.05)。结论:10~100 U/mL浓度的的胎盘异铁蛋白对鼠胚的早期发育没有显著影响,但能促进晚期鼠胚的生长、分化和孵出。 Objective: To assess the effect of placental isoferritin(PLF) in vitro on mouse embryodevelopment. Methods: Two-cell mouse embryos were cocultured with human first trimester decidual cells atdifferent concentrations for PLF in vitro, and the following changes of the above system were closely observedunder invert microscope, at the same time the number of embryos were recorded and the embryos were classified.Results:1) There were no significant difference in the percentage of embryos development to 4-cell, 8-cell andmorula stoges (P>0.05). 2) PLF at the concentrations of 10 U/mL and 100 U/mL significantly enhanced furtherembryos development to the blastocyst and hatching blastocyst (P<0.05). 3) PLF at the concentration of 1 000 U/mL depressed further embryos development from 2-cell to hatching blastocyst, meanwhile such phenomena as celldegeneration and irregular cleavage were observed in part of embryos, but there was no significance in statistics(P>0.05). Conclusions: The results suggested that PLF at the concentration of 10-100 U/mL does not obviouslystimulate early mouse embryo develipment, however, PLF can promote the growth, differentiation, and hatching ofpreimplantion blastocyst.
