人宫颈癌中c-Ha-ras-1癌基因表达的研究

摘要 为探讨宫颈癌中 c-Ha-ras-I 丧达的情况,通过斑点杂交和 Northern 杂交检测了21例宫颈癌,30例宫颈炎,3例正常宫颈组织中 mRNA 的水平,结果表明:c-Ha-ras-I mRNA 在宫颈癌中为81%阳性,重度宫颈炎为70%,中度宫颈炎为40%,轻度宫颈炎为30%,正常宫颈组织勾阴性。薄层扫描表明宫颈癌及重度宫颈炎与其他宫颈组织的 c-Ha-ras-I mRNA 水平有显著性差异(P<0.01).Northern blot 表明:Ha-ras mRNA 为1.4kb。提示:Ha-ras 癌基因的过量表达在宫颈癌的发生发展过程中可能起重要作用. To elucidate the transcription of c-Ha-ras-1 oncogene in cervical carcinoma ,we examined the mRNA level in 21 eases of cervical carcinoma,30 eases of cervieitis,3 cases of normal cervical tissue by dot blot hybridization and Northern blot hybridization.The results showed that the positive per- centage of c-Ha-ras-1 was 81% in cervical carcinoma ,70% in severe cervicitis,40% in middle cer- vicitis,30% in slight cervieitis,and negative in normal tissue.LKB-density scanning technique for quantitively analysizing positive dot showed a significant difference between cervical carcinoma ,se- vere cervicitis and normal cervix,slight middle,severe cervicitis (P<0.01).The Northern blot hy- bridization showed that the mRNA of c-Ha-ras-1 oncogene transcription in cervical carcinoma is 1.4 kb.These results indicated that there was an overexpression of c-Ha-ras-1 in cervical carcinoma and severe cervicitis;the overexpression of c-Ha-ras-1 in cervical tissue is associated closely with the gen- eration and evolation of cervical carcinome.
