脂质过氧化在急性中毒性肾损伤中的作用

The Effect of Lipid Peroxidation on Gentamycin and Mercuric Chloride Induced Acute Nephrotoxicity

摘要 本实验观察大鼠急性庆大霉素和升汞中毒性肾损伤时肾组织中与 LPO 有关指标的变化。结果显示:庆大霉素(60mg/kg ip qd)中毒72h 和升汞(5mg/kg,im)中毒24h 后肾功能受损,肾系数增大;肾组织中的 MDA、FFA 含量增高;XO 活性增高。表明 LPO 增强可能是庆大霉素和升汞中毒性肾损伤的重要介导因素,黄嘌呤-XO 系统激活可能是导致 OFR 产生增多、LPO 增强的重要机制。 Gentamycin (60 mg/kg.ip,q,d3)and mercuric chloride (5 mg/kg im .q.d1)were adminstrated in male Sprague-Dawley rats.The results showed that plasma creatinine and blood urea nitrogen of in- toxicated rat were higher than those of normal control:Malondiyaldehyde,free fatty acid and xan- thine oxidase activities in the toxic kidneys of the test groups were increased singnificantly as compared with those in normal kidneys.These results suggest that lipid peroxidation is involved in the pathogene- sis of gentamycin and mercuric chlorid mediating renal injury,and xanthine-xanthine oxidase may be one major source of oxygen free radicals under such a condition.
