实时荧光聚合酶链反应定量检测准分子激光原位角膜磨镶术后兔角膜转化生长因子β2的表达 被引量:1

摘要 目的 :定量检测转化生长因子 β2 (TGF β2 )在LASIK术后不同时段角膜中的表达水平 ,探讨TGF β2与准分子激光原位角膜磨镶术 (LASIK)术后早期伤口的愈合关系。方法 :取 2 0只新西兰白兔 ,左眼按近视 4D行LASIK术 ,右眼作对照组。分别于术后即刻、4h、4 8h、1w、2w取角膜 ,用荧光半定量PCR方法检测兔中央手术区及周边非手术区角膜TGF β2的表达。 结果 :LASIK术后不同时段术眼中央手术区与周边非手术区角膜TGF β2的mRNA水平差异无显著性 (P>0 .0 5 ) ,左眼手术区与右眼相应对照区角膜TGF β2的表达差异同样无显著性 (P >0 .0 5 )。结论 :LASIK术后反应轻微 ,组织损伤小 ,角膜TGF β2的表达与术前相比无明显变化。 Objective:To quantitatively detect the expression level of the transforming growth factor-β2 gene in the cornea and study corneal wound healing mechanisms in the early period after laser in situ keratomileusis(LASIK).Methods:The left eyes of 20 adult New Zealand rabbits were operated on to produce a myopic ablation of 4.0 diopters. The corneal specimens were prepared at 0,4 h,48 h,and 1 and 2 weeks after surgery. Expression of TGF-β2 was determined by real-time quantitative RT-PCR methods in the central surgical zone and the non-surgical periphery zone.Results:There was no significant difference in the expression level of TGF-β2 mRNA between the central surgical and non-surgical periphery zones. The difference in the expression of TGF-β2 between the surgical zone of the left eye and corresponding area of the right eye was also not significant(P>0.05).Conclusion:There was no significant difference in TGF-β2 expression between normal and post-LASIK corneas. The wound healing response in flap and stromal interfaces was lighter and the cornea remained clear.[
