摘要
目的 :定量检测转化生长因子 β2 (TGF β2 )在LASIK术后不同时段角膜中的表达水平 ,探讨TGF β2与准分子激光原位角膜磨镶术 (LASIK)术后早期伤口的愈合关系。方法 :取 2 0只新西兰白兔 ,左眼按近视 4D行LASIK术 ,右眼作对照组。分别于术后即刻、4h、4 8h、1w、2w取角膜 ,用荧光半定量PCR方法检测兔中央手术区及周边非手术区角膜TGF β2的表达。 结果 :LASIK术后不同时段术眼中央手术区与周边非手术区角膜TGF β2的mRNA水平差异无显著性 (P>0 .0 5 ) ,左眼手术区与右眼相应对照区角膜TGF β2的表达差异同样无显著性 (P >0 .0 5 )。结论 :LASIK术后反应轻微 ,组织损伤小 ,角膜TGF β2的表达与术前相比无明显变化。
Objective:To quantitatively detect the expression level of the transforming growth factor-β2 gene in the cornea and study corneal wound healing mechanisms in the early period after laser in situ keratomileusis(LASIK).Methods:The left eyes of 20 adult New Zealand rabbits were operated on to produce a myopic ablation of 4.0 diopters. The corneal specimens were prepared at 0,4 h,48 h,and 1 and 2 weeks after surgery. Expression of TGF-β2 was determined by real-time quantitative RT-PCR methods in the central surgical zone and the non-surgical periphery zone.Results:There was no significant difference in the expression level of TGF-β2 mRNA between the central surgical and non-surgical periphery zones. The difference in the expression of TGF-β2 between the surgical zone of the left eye and corresponding area of the right eye was also not significant(P>0.05).Conclusion:There was no significant difference in TGF-β2 expression between normal and post-LASIK corneas. The wound healing response in flap and stromal interfaces was lighter and the cornea remained clear.[
