摘要
目的 :观察经内毒素脂多糖 (LPS)刺激的单核细胞培养上清对人牙髓细胞碱性磷酸酶 (ALP)活性的影响 ,探讨单核细胞介导下LPS和CD1 4在人牙髓细胞分化中的作用。方法 :采用经具核梭杆菌LPS刺激的单核细胞培养上清作用于体外培养的人牙髓细胞 ,通过OD值测定 ,观察单核细胞培养上清对人牙髓成纤维细胞碱性磷酸酶 (ALP)活性的影响。结果 :LPS直接刺激人牙髓细胞 ,牙髓细胞ALP活性明显上升 ,但LPS刺激的单核细胞培养上清对人牙髓细胞ALP活性有明显的抑制作用。抗CD1 4抗体在有血清的条件下作用于单核细胞 ,可以阻断单核细胞介导下LPS对牙髓细胞ALP活性的抑制。结论 :单核细胞介导的LPS对牙髓细胞ALP活性有抑制作用 ,其机制可能依赖CD1 4。
Objective:To investigate the influence of Fusobacterum nucleatum (Fn) LPS stimulated monocyte culture supernatant on alkalinephosphatase (ALP) activity of human dental pulp cell,and to explore the effect of LPS mediated monocyte and CD14 on pulp cell differentiation.Methods:The culture supernatant of monocytes stimulated with Fn LPS was applied to human dental pulp cell in vitro,and activity of ALP in human dental pulp fibroblast was monitored by OD test.Results:ALP activity of pulp cell increased after three days of stimulation directly by LPS,while LPS stimulated monocyte culture supernatant markedly inhibited ALP activity.It was found that in the presence of human serum,anti CD14 antibody could act on monocytes and block the inhibition effect of LPS on ALP activity of pulp cell.Conclusion:LPS mediated by monocyte had inhibitory effect on dental pulp cell,which might be CD14 dependent.
