刚地弓形虫RH株GRA2分子基因克隆与序列分析

摘要 目的克隆刚地弓形虫RH株致密颗粒抗原2(Dense Granule Antigen,GKA2)分子基因,为研究使用重组的GKA2分子作为抗原诊断弓形虫病及进行弓形虫分子疫苗研究奠定基础。方法弓形虫DNA的提取。目的基因GKA2片断的扩增、酶切及纯化。目的基因片断的纯化。目的基因的克隆。核酸序列分析。结果从弓形虫RH株基因组DNA中扩增出的402 bp的GRA2基因片断,与以往报道的RH株GKA2分子有100%的同源性。结论本研究成功地获得了刚地弓形虫KH株的GKA2分子基因,为今后此分子作为抗原诊断弓形虫病及进行弓形虫核酸疫苗研究奠定基础。 Cloning of the GRA2 gene from Toxoplasma gondii RH strain,and offering the compound genes of GRA2 for the diagnosis of Toxoplasmosis and the research of Toxoplasma gondii vaccine. The sequence of amplification GRA2 gene fragment was in accord with the reported one matelly. The gene encoding GRA2 was amplified from the DNA of RH strain of Toxoplasma gondii by PCR,which paves way for further study of the diagnosis of Toxoplasmosis and the research of Toxoplasma gondii vaccine.
