摘要
目的 探讨原发上皮性卵巢癌中P15基因mRNA(P15mRNA)及其蛋白的表达情况 ,以确定该基因在原发上皮性卵巢癌发生发展过程中的作用。方法 采用逆转录 聚合酶链反应 (RT PCR)及免疫组化方法 ,对4 8例原发上皮性卵巢癌以及 15例正常卵巢组织P15mRNA及其蛋白进行了检测。结果 4 8例原发上皮性卵巢癌中P15mRNA表达缺失者 11例 (2 2 92 % ) ,P15蛋白缺失者 13例 (2 7 0 8% ,包括 11例P15mRNA表达缺失者 )。15例正常卵巢组织P15mRNA及其蛋白均有表达。P15mRNA及其蛋白表达缺失与原发性上皮性卵巢癌的组织学类型有关 (P <0 0 5 ) ,而与其FIGO分期及组织学分级无关 ,但随肿瘤临床期别和病理级别增高缺失率有增高的趋势。一致性检验显示P15mRNA与其蛋白表达关系密切 (P <0 0 5 )。结论 P15基因在原发上皮性卵巢癌中存在mRNA以及蛋白表达的异常 ,提示该基因的失活与原发上皮性卵巢癌的发生有关。
Objective To study the aberrant expression of P 15 mRNA and protein in primary ovarian epithelial carcinomas,and to define the role of P 15 gene in the generation and development of primary ovarian epithelial carcinomas.Methods By using of reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry assays,expressions of P 15 mRNA and protein were detected in 48 primary epithelial ovarian carcinomas and 15 normal ovarian tissues.Results The loss of expressions of P 15 mRNA and P 15 protein was found to be 11 (22.92%) and 13 (27.08%) respectively.Expressions of P 15 mRNA and P 15 protein were detected in all of the 15 normal tissues.There was a correlation between the expressions of P 15 mRNA P 15 protein and histological types in primary epithelial ovarian carcinomas (P<0.05),and although their expressions were irrelevant to FIGO stages and histological grade,there was a tendency for them to increase with the stage and grade increasing.The expression of P 15 at protein level was significantly correlated with that at mRNA level (P<0.05).Conclusions The aberrant expression of P 15 mRNA and protein suggests that inactivation of P 15 gene is associated with tumorigenesis in primary ovarian epithelial carcinomas.
