摘要
α 转移葡萄糖苷酶是生产低聚异麦芽糖的关键酶。文中探讨了一种黑曲霉所产α 转移葡萄糖苷酶粗酶的分离提纯方法 ,并研究了纯酶的部分酶学特性。该酶可用葡聚糖凝胶G 2 0 0柱层析法分离 ,粗酶液含有多种杂蛋白 ,用SDS 聚丙烯酰胺凝胶电泳测定纯酶的分子质量为 1 3 0ku。该酶既有水解特性 ,又有较强的转移能力。
transglucosidase is a critical enzyme for producing isomaltooligosaccharide. The methods to produce coarse enzyme of α-transglucosidase produced from Aspergillus niger, were studied. The characteristics of the pure enzyme were also studied. The coarse enzyme of α-transglucosidase was obtained using sephadex G-200 column chromatography. the molecular weight of pure enzyme was determined to be 130ku by SDS-PAGE electrophoresis. α-transglucosidase has the properties of both hydrolases and transferases.
