摘要
目的 观察 lac Z报告基因重组腺病毒对人脑胶质瘤细胞 U 2 5 1的转染能力及其细胞毒性。方法 将lac Z报告基因重组腺病毒转染人脑胶质瘤细胞 U 2 5 1,TCID50 法测定病毒滴度 ,MTT法测定细胞毒性影响。结果 腺病毒滴度在 2 .0× 10 6时可使 U2 5 1细胞 10 0 %感染 ;MTT法显示腺病毒转染与未转染的 U2 5 1细胞间 OD570 差异无显著性。结论 针对 U2 5 1细胞而言 ,腺病毒具有很高的转染效率 ,其本身对细胞无毒性 。
Objective To investigate the transfection efficiency and the cytotoxic effects of recombinant adenovirus with lacZ report gene in U251. Methods A recombinant replication-deficiented adenovirus,Adeno-X^(TM)-lacZ, which contains the lacZ report gene, was transfected into U251 cells. TCID (50) assay was used to determine viral titer; MTT assay was used to (evaluate) the cytotoxic effects of Adeno-X^(TM)-lacZ. Results The transfection efficiency reached 100% at 2.0×10~6 pfu/ml; MTT assay showed no significant differences between the OD (570) of transfected and untransfected U251 cells. Conclusion Adenovirus vector is a safe and efficient vector for glioma gene therapy research.
