猕猴桃果实中内源水杨酸的提取、测定及其在采后研究中的应用 被引量:18

Extraction and Determination of Endogenous Salicylic Acid from Kiwifruit and Its Application to Postharvest Research

摘要 建立一种更有效的水杨酸(SA)提取、测定方法是了解SA对采后果实成熟衰老生理影响和调控机理的基础。本文采用乙醚一步萃取法提取猕猴桃果实组织中内源SA,用高效液相色谱法(荧光检测器)测定其含量。结果显示,本方法回收率为(96.4±3.0)%,显著高于其它已报道的方法,最低检测限为0.9ng/g·FW。SA的荧光峰面积与浓度在0~2.0μg/mL范围内呈线性关系,达到极显著水平(r=0.9991),该方法简便易行,样品提取时间比以往报道的方法节省一半,因此可用于成熟果实组织中的内源SA萃取和测定。 The improved method of extracting and determining salicylic acid from fruit tissue is the basis of understanding the physiological effects and regulation mechanism of salicylic acid on fruit ripening and senescence. In this paper endogenous salicylic acid was extracted with ethyl ether from kiwifruit tissue and the concentration was detected by High-performance Liquid Chromatography with fluorescence detector. The results showed that the recovery of the method was (96.4±3.0)% and higher than that of other reported methods the lowest limit of determination was 0.9 ng/g·FW and the linear range of the method was 0~2.0μg/mL(r=0.9991). The method was simpler and easier than that of others and the extraction time was half of other reported methods. The desirable result could be obtained by using this improved method for extracting and determining endogeous salicylic acid from ripening fruit tissue.
