摘要
基于不同类型抑制剂存在的酶催化反应的特点 ,利用计算机编程判定抑制剂类型和求定动力学参数。处理时先对实验测定的不同底物 Cs分别在不存在和存在抑制时对应的反应速率 V0 和 v,利用线性回归法求出 1/V0 ~ 1/Cs和 1/v~ 1/Cs的关系模型 ,然后通过比较两直线方程的斜率和截距以及交点坐标的特点 ,判定抑制剂类型 ,最后根据不同类型抑制剂的动力学方向求出动力学参数。文中以羧肽酶 A催化底物 Bz-G1y-G1y-Phc分解反应和富马酸水解成苹果酸的反应实验测定结果为例进行验证 ,结果表明 ,利用所编程序确定的抑制剂类型与常规方法很好的吻合。在酶催化动力学研究中利用这种数据处理方法将使实验过程得到简化 ,实验结果更准确、更可靠。在数据处理过程中 ,为高效、快速确定抑制剂类型和求定动力学参数 ,采用了开放式计算机编程语言 Matlab进行程序设计 ,并在文中提供了程序设计流程和实用源程序清单。
Based on the characteristics of enzymic catalytic reaction,the paper introduces the method of determining the inhibitor type and the kinetic parameters in enzymic catalytic reaction by designing computer program.During the process,for the measured substrate value Cs and the reaction rate V 0 and v at the differential conditions of existential inhibitor or not,the linear regression method isused to obtain the relationships of 1/V 0~1/Cs and 1/v^1/Cs,and then to determine the inhibitor type by comparing the feature and the cross point of the two linear equation,at last to obtain the kinetic parameters by using the kinetic equation of various inhibitor.Calculation examples for experimental data of the decomposition reaction of protaminase at the catalytic substrate of Bz-Gly-Gly-Phc and the hydrolytic reaction of allomaletic acid are given,the results show a good agreement to the results computed with ordinary method.In the research of enzymic catalytic kinetics,the analytic method of experimental data processing will simplify the experimental procedure,and the results may be more exact and reliable.Language Matlab is applied as program language in order to keep the program simple,short and efficient,and the flowchart of run course and the program list are too listed in the paper.
