摘要
目的 探讨大鼠局灶性脑缺血再灌注后各时相点海马CA1区超微结构以及中药川芎嗪对该区影响。方法 采用闭塞大鼠大脑中动脉建立局灶性脑缺血再灌注模型 ,透射电镜下观察海马CAI区超微结构改变。结果 实验组海马CA1区超微结构的改变总体较对照组明显减轻 ,表现为 :再灌注 6h (B1组 )的星形胶质细胞 ,突起稍肿胀 ,线粒体结构无明显变化 ;神经细胞内仅部分线粒体肿胀 ,内质网扩张。再灌注 12h (B2组 )的星形胶质细胞 ,突起肿胀 ,线粒体结构无明显变化 ;未与星形胶质细胞相连的神经细胞 ,胞浆内线粒体肿胀、轴突内线粒体空泡化 ;与星形胶质细胞相连的神经细胞 ,内质网脱颗粒 ,线粒体无明显改变。结论 川芎嗪不仅直接保护脑神经元 ,而且通过保护星形胶质细胞而间接起到保护和修复受损神经细胞的作用。
Objective To investigate the ultrastructure changes and the effect of ligustrazine in hippocampi CA1 area after local cerebral ischemia reperfusion.Methods Rat middle cerebral artery occlusion/reperfusion (MCAO) models were adopted,and at different time points the brains were removed.The electron microscope was applied to observe the ultrastructure changes of hippocampi CA1 area and the effect of ligustrazine.Results In the experimental group (ligustrazine treatment),the ultrastructure of CA1 area lightened more obviously than that in the control group,which showed that neurons and astrocytes at reperfusing 6h bulged and expanded,mitochondrion structure had no obvious change;in verve cells,only some mitochondria expanded,endoplasmic reticulum bulged.Astrocytes at reperfusing 12 h bulged and expanded;mitochondrion structure had no obvious change.In nerve cells which were not linked with astrocytes,mitochondria in cytoplasm expanded,mitochondria in axon vacuoled;in nerve cells which were linked with astrocytes,endoplasmic reticulum degranulated,and mitochondrion structure had no obvious change.Conclusion Ligustrazine can not only protect the neuron and astrocyte at one time,but also has the indirect protection and restoration on the damaged nerves.
