摘要
以MS固体培养基为基本培养基,研究了盾叶薯蓣(DioscoreazingiberensisC.H.Wright)细胞在不同接种量和NAA,BA浓度配比条件下的生长特征。结果表明:盾叶薯蓣细胞在MS培养基,附加NAA2 0mg/L,BA0 5mg/L,蔗糖30g/L,琼脂7g/L,pH6 0,接种量0 8~1 2g/瓶条件下,生长较好。应用Logistic方程对薯蓣细胞生长进行模拟能够较好地描述细胞的生长过程,细胞生长曲线呈"S"形,生长过程可分为延迟期、指数生长期和减速期。为发酵培养盾叶薯蓣细胞和研究其次生代谢产物提供了理论依据。
Effects of inoculum size and concentration ratio of NAA and BA on the growth of Dioscorea zingiberensis C.H.Wright cells were studied. The results showed that MS supplemented with NAA 2.0 mg/L and BA 0.5 mg/L was the optimum condition for cell culture. The optimum inoculum size was 0.8~1.2 gFW/flask. The growth curve could be described with Logistic function and was 'S' shape. The growth cycle of Dioscorea was divided into three phases: lag phase, exponential phase and decelerating phase.
