新生大鼠视网膜新生血管模型的制备

摘要 目的 建 立大 鼠 视 网 膜新 生 血 管 模型 进 行 大 鼠视 网 膜 新 生血 管 模 型 的 可行 性 研 究 ,并 探 讨 视网膜新 生 血管 的发 病 机制 。方 法 7 d 龄有 色大 鼠 (Brown Norway rat)在浓 度为 75% 的高 氧 环 境中 生 活 5 d,然 后回到正 常 氧环 境 中。 通 过 相 对分 子 质 量 高的 荧 光 素 造影 视 网 膜 铺片 和 视 网 膜组 织 病 理 切 片检 查 ,反映 视 网 膜 血管的增 生 ;利 用分 子生 物 学技 术蛋 白 质印 迹法 (W estern blot)测定 视 网膜 血管 内 皮生 长因 子 (V EGF)含 量的 变 化。结果 给 氧组 新生 有 色 大鼠 视 网 膜 大血 管 显 著 扩张 ,大 片无 灌 注 区 ,大 量 新生 血 管 芽 形成 ;大 量 新生 血 管 突 破视 网膜 内界 膜 长 入 玻璃 体 腔 ;视 网 膜 VEG F 的 表 达 明 显增 强 。 结 论 新 生 有 色大 鼠 可 以 作为 视 网 膜 新生 血 管 模 型的实验 用 鼠。 Objective To establish a model of retinal neovascularization in the rat with reproducible and quantifiable proliferative retinal neovascularization. Methods Twenty of one-week-old pigmented Brown Norway rats were exposed to 75% oxygen for 5 days and then to room air, and another 20 rats of the same age kept in room air as controls.A novel fluorescein-dextran perfusion method has been developed to assess the vascular pattern; The retinal neovascular response was quantitated on sagittal cross sections; vascular endothelial growth factor (VEGF) expression in the retina was determined by Western blot analysis. Results Fluorescein-dextran angiography delineated the entire vascular pattern, including neovascular tufts and large areas of nonperfusion in flat-mounted retinas in oxygen-induced retinopathy in pigmented brown Norway rats and the proliferated vessels extending from the retina into the vitreous cavity on sagittal cross sections. Western blot analysis showed that VEGF expression was significantly increased in the retina by the oxygen treatment. Conclusion A reproducible and quantifiable rat model of oxygen-induced retinal neovascularization developed by the author proved useful for the study of pathogenesis of retinal neovascularization.
