丙型肝炎病毒核心蛋白、Bcl-2、P21^(waf1)在丙型肝炎中的表达及相互关系

摘要 目的 调查丙型肝炎、肝硬化病人组织中的丙型肝炎病毒 (HCV)核心蛋白、P2 1、Bcl 2的表达以及相互间关系 ,探讨HCV核心蛋白是否抑制P2 1和促进Bcl 2表达。方法 收集 2 3例HCV表达阳性的肝炎、肝硬化组织 ,采用免疫组化Envision法检测核心蛋白、P2 1、Bcl 2表达 ,并用统计学方法分析三者间的表达关系。结果 HCV核心蛋白和突变P2 1的阳性表达主要位于细胞核中 ,Bcl 2的阳性表达主要位于细胞质 ;HCV核心蛋白表达阳性组织中 ,P2 1阳性表达率仅 13 % ,Bcl 2阳性表达率则达 95 .7% ,三组间比较差异有显著性 (P =0 .0 12 )。HCV核心蛋白和Bcl 2二组间比较差异无显著性 (P =0 .5 61) ;HCV核心蛋白与Bcl 2、p2 1阳性强度两者间P值分别为 0 .0 12和 0 .2。结论 三种蛋白的表达有相关性 ,HCV核心蛋白可能促进Bcl 2蛋白的表达 ,抑制P2 Objective To investigate expressions of hepatitis C virus (HCV) core,P21 and Bcl 2 proteins in the tissues of HCV infected patients. Methods Liver tissues of 23 HCV infected patients and the expression of HCV core, P21, Bcl 2 proteins were determined with Envision immunohistochemical method, the correlation of the three proteins was analyzed. Results The positive expression of HCV core and mutation P21 protein was primarily in nucleus, while the positive expression of Bcl 2 protein was in cytoplasm. In HCV core positive expressed liver tissues, the positive rate of P21 was 13% and the positive rate of Bcl 2 was 95.7%. The difference was significant ( P = 0.000 ) among the three protein expressions, but there was no difference ( P = 0.561 )between HCV core and Bcl 2. P value of positive intensity between HCV core and p21, HCV core and Bcl 2 was 0.2 and 0.012 , respectively, the correlation coefficient was 0.4 and 0.72 , respectively. Conclusion The expressions of the three proteins is of correlation. It is possible that HCV core protein promotes the expression of Bcl 2 protein and reduces the expression of P21 protein.
