摘要
目的 :研究汉防己甲素 (Tet)对成纤维细胞增殖和胶原合成的作用 ,及其与一氧化氮 (NO )的关系。方法 :体外培养人胚肺成纤维细胞 (HLF) ,应用3H_TdR及3H_脯氨酸参入法观察Tet对HLF细胞增殖及胶原合成的作用 ;应用比色法测定细胞培养液NO水平及一氧化氮合酶 (NOS)活性。结果 :与对照组比较 ,Tet能明显降低3H_TdR及3H_脯氨酸参入率 ;同时 ,明显升高细胞培养液NO水平及NOS活性。结论 :Tet具有抑制成纤维细胞增殖及胶原合成作用 ,NOS活性升高 ,NO合成、释放增加是Tet抑制成纤维细胞增殖及胶原合成作用机制之一。
Objective:To study the effects of tetrandrine(Tet)on fibroblast multiplication and collagen stro-ma synthesis and its relation with NO.Methods:Human embroyo lung fibroblast(HLF)was cultured in vitro. 3 H-TdR incorporation and 3 H-Proline endosmosis methods were used to observe the role of Tet on fibrolast multiplication and collagen stroma synthesis.The method of colorimetry was adopted to investigate NO level and NOS activity of cell culture fluid.Results:Tet could significantly decrease 3 H-TdR, 3 H-Proline endosmosic rate and increase NO level and the NOS activity of cell culture fluid compared with the normal group.Conclu-sion:Tet can inhibit fibroblastic multiplication and collagen strome synthesis.The increase of NO synthesis and release,and NOS activity are one of the mechanisms for Tet inhibiting fibroblast multiplication and collagen strome synthesis.
