阿尔茨海默病患者皮质额叶基因表达谱研究

Genic expressive spectrum of the frontal cortex in patients with Alzheimer disease

目的:通过研究阿尔茨海默病(Alzheimerdisease,AD)患者皮质额叶中发生差异表达的基因,揭示AD发生、发展的分子基础。方法:利用基因芯片分析3例AD患者皮质额叶中基因表达谱,通过与4例正常对照间的比对,寻找存在表达差异的基因。结果:所检测的5000个基因中,共有49个基因存在着差异表达。在患者中13个基因表达水平升高,36个基因表达水平降低。上述基因与多种生理过程,如氧化应激、胆固醇平衡、钙信号转导、炎症反应等密切相关。结论:AD的发生、发展涉及多种基因表达水平和多个生理过程的改变。

AIM:According to genic expressive spectrum of the frontal cortex in patients with Alzheimer disease(AD),the molecular elements of the occurrence and the development in AD are explored. METHODS:Genic chip was used to analyze genic expressive spectrum of the frontal cortex in three AD patients,which aimed to find out genes with different expression compared with four normal patients. RESULTS:A total of 49 genes had significant difference among 5 000. Thirteen genic expressions were increased,and 36 were decreased in the patients.Above all,the genes were closely related to many physiological functions,such as oxidative stress, cholesterol homeostasis,calcium signal transduction, inflammation response,etc. CONCLUSION:The occurrence and development of AD are related to the changes of multi genic expressive levels and many physiological functions.
