摘要
目的:探讨电针预防庆大霉素对耳蜗螺旋神经节细胞(cochlearspiralganglioncells,SGC)损伤的作用。方法:选用耳郭反射正常,体质量为300~350g的健康杂色豚鼠,雌雄兼用,随机分成3组:对照组10只,肌肉注射生理盐水2mL/(kg·d);庆大霉素组15只,肌肉注射硫酸庆大霉素80mg/(kg·d);电针组15只,给予电针(针刺双侧“听宫、翳风、肾俞”穴)1次/d;针刺后再给予庆大霉素同对照组,各组用药均25d。用脑干听觉诱发电位(BAEP)检测动物BAEP反应阈值;用组织学方法检测耳蜗SGC的形态学变化。结果:在用药前对照组组、庆大霉素组和电针组BAEP反应阈分别为:(31.85±2.19),(32.23±2.26),(32.19±2.24)dBpeSLP;用药后27d检测分别为:(32.27±2.21),(59.58±17.74),(45.67±9.35)dBpeSLP。庆大霉素组、电针组分别与对照组组比较,差异均有显著性意义(t=6.809,4.435,P<0.01);庆大霉素与电针组比较差异亦有显著性意义(t=3.800,P<0.01)。在放大倍数为400倍光镜下平均在80μm×80μm视野内SGC存活数为:对照组(14.08±0.79)个,庆大霉素组(9.16±2.55)个,电针组(11.17±2.08)个;庆大霉素组与电针组比较差异有显著性意义(t=2.367,P<0.05)。结论:电针对庆大霉素引起的耳蜗螺旋神经节细胞损伤有明显的预防作用。
AIM:To investigate the effects of electroacupuncture(EA) protection on the injury of gentamicin(GM) induced cochlear spiral ganglion cells(SGC). METHODS:Healthy mottle guinea pigs with normal reflexes of pinna(300 g to 350 g in body mass, male or female) were selected and randomly divided into 3 groups: 10 in control group(CN),intramuscularly injected with physiological saline 2 mL/kg per day;15 in GM group, intramuscularly injected with GM 80 mg/kg per day;15 in EA group were given EA at tinggong(SI19),yifeng(SJ17) and shenyu(UB23) point once a day intramuscularly.The duration of administration was 25 days for GM group and control group after EA.The reaction threshold of animal brainstem auditory evoked potential(BAEP) was examined by BAEP.The morphological changes of SGC were examined morphologically. RESULTS:BAEP reaction thresholds of CN,GM and EA groups before administration were(31.85±2.19),(32.23±2.26) and(32.19±2.24) dB peSLP;BAEP reaction thresholds of CN,GM and EA groups 27 days after administration were(32.27±2.21),(59.58±17.74) and(45.67±9.35) dB peSLP,respectively. There was a significant difference(t= 6.809, 4.435,P< 0.01) between CN group and GM group,and between CN group and EA group. There was a significant difference(t=3.800,P< 0.01) between GM and EA groups.The average number of survival SGC in the NC,GM and EA groups in a visual field(80 μm×80 μm) under light microscope(400 times) was 14.08±0.79,9.16±2.55 and 11.17±2.08,respectively. There was a significant difference(t =2.367,P< 0.05) between GM and EA groups. CONCLUSION:EA can prevent the injury of cochlear SGC induced by GM.
