骨髓间充质干细胞移植对缺血再灌注损伤肾组织细胞增殖的影响 被引量:2

摘要 目的探讨外源性的骨髓间充质干细胞(mesenchymalstemcells,MSCs)对缺血再灌注肾损伤的肾组织细胞增殖的影响。方法取SD雄性大鼠胫、股骨骨髓,percoll密度梯度分离法分离骨髓MSCs。32只雌性SD大鼠复制缺血再灌注肾损伤的模型,随机分为两个组:移植组和对照组。移植组经下腔静脉移植骨髓MSCs,对照组给予等量生理盐水,分别于1天、2天、3天、4天处死大鼠,两组大鼠每天均经腹腔注射5-溴脱氧尿苷嘧啶(bromodeoxyuridine,BrdU)。留取肾组织采用免疫组织化学染色方法检测BrdU阳性细胞在肾组织内的分布情况。结果在缺血再灌注后的第二天,移植组和对照组都有表达BrdU阳性细胞,并随时间延长BrdU阳性细胞个数逐渐增多,但移植组BrdU阳性细胞明显高于对照组,两者有统计学意义(P值<0.05)。结论移植的外源性骨髓MSCs能够促进损伤肾脏的细胞增殖,参与肾损伤的修复。 Objective To study the effect of bone marrow mesenchymal stem cells(MSCs) on cells proliferation in renal tissues with ischemia-reperfusion injury in rats. Methods Bone marrow mesenchymal stem cells were collected from femur and tibia of male SD rat, then isolated by percoll density gradient centrifugation. 32 fermale SD rats were divided randomly into the transplanted group and the control group. All rats were subjected to clamping renal pedicles for 45 minutes, followed by injection of MSCs or saline into the inferior vena cava. All animals received daily injection of bromodeoxyuridine(BrdU;50mg/kg,i.p.) for identification of newly synthesized DNA. Animals were sacrificed at 1,2,3,4 days after operation, respectively. Immunohistochemistry were performed to identify BrdU in renal tissues. Results BrdU-positive cells were found in renal tissues in two groups at the 2nd, 3rd and 4th days, and the BrdU-positive cells in transplanted group rats were much more than that of the control group rats(P<0.05). Conclusion Transplanted extra-renal bone marrow MSCs can promote cells proliferation in renal tissues with ischemia-reperfusion injury.
