摘要
目的 :观察槟榔提取物对口腔黏膜角朊细胞分泌肿瘤坏死因子α的影响 ,探讨槟榔诱发口腔黏膜下纤维性变的机制。方法 :通过体外培养口腔黏膜角朊细胞 ,用不同浓度槟榔提取物对其进行干预 ,放射免疫方法检测不同干预时段培养上清中的肿瘤坏死因子α浓度。结果 :槟榔提取物干预角朊细胞 12h后肿瘤坏死因子α的量开始增加 ;并以剂量依赖关系增加角朊细胞产生肿瘤坏死因子α的量。结论 :槟榔提取物可能通过促进角朊细胞合成肿瘤坏死因子α诱发口腔黏膜下纤维性变。
Objective:To explore the pathogenesis of oral submucous fibrosis(OSF) by observing the effect of Aqueous areca nut extract(AANE) on the level of tumor necrosis factor α(TNFα) produced by oral keratinocytes.Method:Human oral mucosa keratinocytes were isolated ,cultured, stimulated with AANE, The supernatant was aspirated and the level of TNFα was detected using radioimmunoassay. Result :The concentration of TNFα increased after keratinocytes were stimulated by AANE for 12 hours or longer time.AANE can increase the concentration of TNFα secreted by keratinocytes in dose-dependent relationship.Conclusion:Areca nut maybe induced OSF by increasing the level of TNFα produced by keratinocytes
