JY97-1×Glenlia杂种F_4代贮藏蛋白的遗传分析

摘要 为了解小麦JY97-1×Glenlia杂种F4代贮藏蛋白的遗传现象,采用SDS-PAGE法和APAGE法分析了JY97-1×Glenlia杂种F4代100个单株的高分子量麦谷蛋白亚基(HMW-GS)组成和醇溶蛋白遗传多样性。结果表明,(1)F4代共出现20种HMW-GS组成,Glu-A1、Glu-B1、Glu-D1三个位点上分别检测到2、8、7种不同的亚基类型。Glu-B1位点出现20、6+8、7、8等六种新类型;Glu-D1位点出现2+12、5+10、5+12等六种新的类型。优质亚基组合1,7+8,5+10的比例为13.71%。(2)Glu-B1b及Glu-B1c基因的不完全表达率达2.11%。(3)醇溶蛋白谱带变异形式有29种,每种带型的谱带数变幅在14~26之间,平均为21.38条。ω、γ、β、α区分别有22、19、7、11种变异,表明Gli-1变异最丰富。聚类结果表明,后代与母本聚为一类的类型最多,表明后代醇溶蛋白带的遗传有偏母现象。 For finding out genetic law of wheat storage proteins, SDS-PAGE and APAGE analysis were used to analyse the (variations) of high molecular weight glutenin subunit compositions and gliadin in F_4 of JY97-1×Glenlia. The (results) showed that there were 20 different (HMW-GS) composition types in F_4 lines. 2, 8 and 7 types of HMW-GS types were found in Glu-A1, Glu-B1 and Glu-D1, respectively. At Glu-B1 loci, there were six new subunit: 6+8, 20, 7, 8 etc. At Glu-D1 loci, there were 2+12,5+10; 5+12 etc six new subunit types. The rate of the best composition of high molecular weight glutenin subunit (1,7+8,5+10) was (13.71%). The incomplete expression rate of Glu-B1b and Glu-B1c was 2.11%. 29 different gliadin (patterns) were found, each patterns had 14 to 26 bands, the average was 21.38 bands. There were 22, 19, 7, 11 kinds of (variation) in the zone of ω,γ,β and α, which showed the loci of Gli-1 possessed more extensive (variation). Cluster analysis showed that the female cluster had the most gliadin patterns.
