粟酒裂殖酵母一个新的box C/DsnoRNA的鉴定、功能分析及进化意义 被引量:2

摘要 通过筛选粟酒裂殖酵母核内小分子RNA的cDNA文库, 获得了一个新的小分子RNA序列. 该RNA具有典型的box C/D snoRNA的保守元件和结构特征, 并具有一段与25S rRNA互补的、长度为10个核苷酸的反义序列, 推测其可指导25S rRNA G940 位点2'-O-核糖甲基化修饰. 经dNTP浓度依赖性引物延伸实验验证, G940确实为2'-O-核糖甲基化修饰位点. 功能元件比较分析揭示, 该snoRNA的上游反义序列与酿酒酵母snR60 的下游反义序列同源, 故命名为snR60-Ⅱ. snR60-Ⅱ基因位于一个非编码RNA基因的内含子中, 其产物由宿主基因转录后的RNA前体加工而来. 通过计算机分析, 从真菌和果蝇中分别发现了一批酵母snR60功能同源分子. 在不同生物中, snR60存在独立基因编码、内含子编码和多顺反子转录等多种基因组织形式, 表明snR60在进化过程中具有高度的移动性.
