摘要
目的 :观察SBHL对HeLa细胞生长的抑制作用 ,并探讨其作用机制 .方法 :用MTT比色法观察不同浓度SBHL对HeLa细胞增殖的影响 ;用流式细胞术对细胞周期进行分析 ;用免疫组化方法观察SBHL对HeLa细胞人端粒酶逆转录酶 (humantelomerasereversetranscriptase,hTRT)基因表达的影响 .用瑞氏 姬姆萨染色法观察SBHL对HeLa细胞形态的影响 .结果 :0 .1~ 2 0 .0mg/LSBHL对HeLa细胞增殖有明显的抑制作用 ,这种抑制作用随SBHL浓度的增加而增加 ;流式细胞术对细胞周期进行分析的结果显示 ,随着SBHL浓度的增加 ,S期细胞含量逐渐降低 ,G1期细胞含量逐渐增加 ,SBHL将细胞阻滞于G1期 ,抑制DNA合成 .SBHL处理的HeLa细胞与对照组相比 ,hTRT基因的表达明显降低 ;瑞氏 姬姆萨染色结果表明SBHL可使HeLa细胞向良性分化 .结论 :SBHL明显抑制HeLa细胞生长呈浓度依赖性 .SBHL下调HeLa细胞hTRT基因的表达 ,抑制端粒酶活性 .
AIM: To observe the inhibiting effects of SBHL on the growth of HeLa cells and to investigate the inhibiting mechanisms. METHODS: The proliferation of HeLa cells was observed by MTT assay and the changes of the cell cycle of HeLa cells treated with SBHL were analyzed by flow cytometry. The expressions of hTRT gene were detected by immunohistochemistry techniques and the differentiation of HeLa cells was observed by Wright-Giemsa staining. RESULTS: The proliferation of HeLa cells was inhibited at 0.1-20.0 mg/L SBHL. The inhibiting effect increased gradually following the increase of concentration of SBHL. The percentage of HeLa cells in S phase decreased and the percentage of cells in G1 phase increased after the cells were treated by SBHL, indicating that SBHL arrested HeLa cells in G1 phase and thus the synthesis of DNA was inhibited. After treatment by SBHL, the expressions of hTRT gene of HeLa cells decreased obviously. Differentiated HeLa cells wwere found after treatment by SBHL. CONCLUSION: ① The growth of HeLa cells is inhibited obviously by SBHL and the inhibiting effect is dependent on the concentration of SBHL. ② The expressions of hTRT gene are downregulated, the telomerase activity is inhibited and the growth of HeLa cell is inhibited.
