摘要
目的 :改进检测人类性染色质的方法 .方法 :以人类口腔黏膜细胞为材料 ,改进离心滴片法操作流程 ,改变三种染液硫堇、吉姆萨、冰醋酸龙胆紫的浓度 ,用扣染法进行染色 .结果 :改进后的检测效果好 ,三种染液硫堇、吉姆萨、冰醋酸龙胆紫所染出的性染色质出现率均等 ,分别为 (2 0 .3± 1.4 ) % ,(19.5± 1.5 ) % ,(2 0 .0± 1.0 ) % .结论 :应用改进的人类性染色质检测方法 ,可以准确检出性染色质 .
AIM: To improve the detection method of human sex chromatin. METHODS: Human buccal mucosal cells were used as materials, the operation of centrifugal dropping slide was improved and the concentration levels were changed in three dyes, Thionin, Giemsa and Acetic-gentian violet (AGV). Cover dyed method was employed in staining. RESULTS: Equalized Barr Body Frequency (BBF) was obtained in the samples that had been dyed respectively by Thionin, Giemsa and Acetic-gentian violet (20.3±1.4)%, (19.5±1.5)% and (20.0±1.0)% respectively. CONCLUSION: The application of the improved detecting method of human sex chromatin enables us to detect sex chromatin immaculately.
