摘要
目的 :研究在轻度修饰低密度脂蛋白 (mm LDL)诱导下差异表达的基因 ,探讨动脉粥样硬化发生的分子机制。方法 :用DDRT PCR(differential displayreversetranscription PCR)技术分析mm LDL诱导下人血管内皮细胞的基因表达差异 ,并用反向Northern证实差异显示基因。结果 :mm LDL诱导下人血管内皮细胞出现一些上调和下调的基因 ,上调基因有胸腺素β4、FGFRI原癌基因伴随蛋白、FK5 0 6结合蛋白、rTSβ蛋白和细胞间粘附分子 1 ;下调基因有Apobec 1结合蛋白 1、细胞色素B5 61和ERP72。结论 :体外mm LDL可诱导HUVEC细胞一些基因表达水平变化 。
Objective: To study the differences of genes' expression in vascular endothelial cells induced by mildly modified low density lipoprotein (mm-LDL) and to help understand the molecular mechanism of atherosclerosis. Methods: Differential-display reverse transcription-PCR (DDRT-PCR) was used to analyse the differences of genes' expression in vascular endothelial cells induced by mm-LDL. The differential expressed cDNA was confirmed by reverse Northern blot. Results: The cDNAs of up-regulated and down-regulated genes were isolated in HUVEC induced by mm-LDL. The up-regulated genes included human thymosin β4, FGFRI oncogene partner, FK506 binding protein, rTSβ protein and intercellular adhesion molecular-1 were identified. The down-regulated genes included human Apo becl binding protein 1, cytochrome B561 and ERP72 were identified. Conclutions: Mm-LDL could change the expression of some genes in endothelial cells and result in the pathological changes. Finally, the formation of atherosclerosis speckle could be related to these genes.;
