饮酒对运动员机体伤害的机制研究:不同浓度乙醇对小鼠十二指肠黏膜上皮细胞的影响 被引量:2

摘要 目的:观察饮用不同浓度乙醇实验小鼠十二指肠中段上皮细胞凋亡情况,探讨饮酒作为一种不良刺激对细胞凋亡究竟有何影响。方法:实验小鼠随机分成4组,每组9只。正常对照组,饮用自来水;50mL/L乙醇组,饮用50mL/L乙醇水溶液;100mL/L乙醇组,饮用100mL/L乙醇水溶液;150mL/L乙醇组,饮用150mL/L乙醇水溶液。各组每日喂食前先将饲料烘干。每天更换新配制的乙醇溶液1次。从22d龄幼鼠开始,同时饲以不同浓度的乙醇,连续喂养60d实验结束。脊髓脱臼法处死小鼠,然后提取十二指肠中段上皮细胞DNA,经琼脂糖凝胶电泳来检测细胞凋亡情况。结果:实验小鼠十二指肠黏膜上皮细胞DNA均有不同程度的损伤,经琼脂糖凝胶电泳后,在胶板上得到不同的梯形带,呈现细胞凋亡的基本特征,对照组:梯形带不明显;50mL/L乙醇组:梯形带明显;100mL/L乙醇组:梯形带较明显;150mL/L乙醇组:梯形带明显。结论:乙醇能明显引起实验动物非正常细胞凋亡。 AIM:To observe the apoptosis of epithelial cells in the middle duodenum of mice after drinking alcohol of different concentrations,and explore the effects on apoptosis resulting from drinking as an unhealthy stimulus. METHODS:Experimental mice were divided randomly into 4 groups with 9 mice in each group:control group(drinking tap water),50 mL/L alcohol group(drinking 50 mL/L alcohol solution),100 alcohol mL/L group(drinking 100 mL/L alcohol solution) and 150 ml/L alcohol group(drinking 150 mL/L alcohol solution).The feed was dried before feeding in each group every day,the fresh alcohol solution was changed every day.The 22 day old mice were fed by alcohol of different concentrations for successive 60 days.The mice were killed by spinal cord dislocation when the experiment was over,then the DNA in epithelial cells of middle duodenum was extract,and the apoptosis of DNA was determined with agarose gel electrophoresis. RESULTS:There were different degrees of DNA damages in epithelial cells of middle duodenum.Different ladders appeared after agarose gel electrophoresis,which revealed the basic characteristics of apoptosis.The ladder was not clear in the control group,but clear in the 50 mL/L and 150 mL/L alcohol groups,and clearer in the 100 mL/L group. CONCLUSION:Alcohol obviously leads to abnormal apoptosis in experimental animals.
