人神经营养素-4成熟蛋白基因克隆 被引量:1

摘要 本研究的目的是克隆人神经营养素-4成熟蛋白基因(mhNT-4)并进行序列分析。应用PCR技术,以正常人血淋巴细胞染色体DNA为模板,扩增出人神经营养素-4(hNT-4)成熟蛋白编码基因。将所得基因片段重组于pGEM-T Easy质粒,筛选得到含人mhNT-4基因的阳性克隆。采用Sanger单链末端终止法测出全部的核苷酸序列。所得到的序列与国外文献所报道的结果(GenBank,M86528)完全相同。mhNT-4成熟蛋白基因的成功克隆,为研究其在原核细胞中的表达提供了有利条件。本文结果也提示不同人种间及不同个体间hNT-4成熟蛋白基因是相对保守的。 Our purpose is to clone and sequence the matured human neurotrophin-4 (mhNT-4) gene. With the chromosomal DNA of human blood lymphocytes as template, mhNT-4 coding genes were amplified by polymerase chain reaction(PCR) and re combinated into phage vector pGEM-T Easy, which was sequenced by using Sanger's single stranded DNA terminal termination method. The sequence of the cloned gene was completely the same as that reported in the literature (GenBank data base, M86528). This study successfully cloned and sequenced the gene of mhNT-4, and it would be convenient to study the expression of mhNT-4 in prokaryocyte. This study indicates that the matured hNT-4 is conservative in different races and individuals.
