山溪鲵皮肤分泌物抗菌活性的初步研究 被引量:12

摘要 山溪鲵皮肤分泌物和经两次SephadexG 2 5层析分离得到的初步提纯样品经抗菌实验 ,发现具有抗菌活性。其中的抗菌活性成分为蛋白质性质 ,分子量约为 2 0kD ,具有在高温下短时间、常温下长时间保持稳定的特性。初步提纯样品经尿素SDS聚丙烯酰胺凝胶电泳检测为两条带 ;经反相高效液相层析 (RP HPLC)检测为 13个峰。此提纯方法的活性回收率为 4 6 .7%。这为进一步的纯化和性质鉴定工作奠定了基础。 The skin secretion of Batrachuperus pinchonii and the Part Purified Substance,acquired by the skin secretion loaded on Sephadex G-25 chromatography column twice,have antimicrobial ability by antimicrobial test.The antimicrobial active substance is protein property,about 20 KD,stable in high temperature during short time and in normal temperature during long time.By electrophoresis in urea polyacrylamide gel with sodium dodecyl sulftate,the Part Purified Substance has two bands;by RP-HPLC chromatograph,it has thirteen peaks.The recovery of activity is 46.7%.On the basis of which further works including final refining and determining of the active substance can be finished.
