摘要
采用多级提取的方式从菊科植物芙蓉菊枝和叶中分离得到芙蓉菊多糖 ,脱色、除蛋白后 ,部分酸水解该多糖 ,将水解得到的寡糖经过活性炭柱层析得到四个洗脱组分。然后应用糖尿病药物筛选模型 ,考察这四个洗脱组分对于小鼠胰岛组织的胰岛素分泌水平的影响。正常胰岛细胞分泌中空白组胰岛素含量为 7.6 94 ;芙蓉菊寡糖水洗脱组分的胰岛素含量为 8.5 0 6 ;5 %乙醇洗脱组分的胰岛素含量为 9.6 89。结果表明 ,芙蓉菊寡糖水、5 %乙醇洗脱组分有一定的促进胰岛素分泌的作用。
One kind of polysaccharide was isolated from the dried Chinese herbs—Crossostephium chinense by fractional preparation,after decolorization an eliminating protein,it was hydrolyzed by acid incompletely.The activated carbo column chromatography was used to separate this and four kinds of oligosaccharid extracts were obtained by different elution liquid.Using cell-based model for dru screening,test the level of insulin secretion promoted by these four oligosaccharid extracts.The blank insulin secretion of normal islets was 7.694,oligosaccharid aqueous extract was 8.506 and oligosaccharide 5% ethanol extract was 9.689.The results indicated that Crossostephium chinense oligosaccharide aqueous extract an 5% ethanol extract could promote insulin secretion.
