米托蒽醌与维拉帕米合用对ACC—2细胞周期运行的影响

摘要 应用流式细胞计(FCM)研究了经抗癌药物米托蒽醌(DHAD)和钙拮抗剂(VP)单独或联合处理的人唾液腺腺样囊性癌 ACC—2细胞的周期运行情况。发现:VP 在20μg/ml浓度范围内对ACC—2细胞增殖周期无明显影响。DHAD 使细胞阻滞在 G_2M 期,并呈剂量依赖性;当剂量一定时,随时间延长,对G_2M 期阻断作用减轻,细胞周期运行有恢复趋向。DHAD 佐以 VP 则明显增强对 ACC—2细胞 G_2M 期的阻断作用。本实验结果为阐明 VP 增强 DHAD 抗癌作用的机制提供了依据。 In flow cytometric studies,the authors investigated the effects of VP and DHAD on ACC—2 cell cycle tra-verse.The authors found that nontoxic concentrations of VP had no block effect on cell cycle traverse.DHAD,in adose-dependent manner,caused ACC-2 cell accumulation in the G_2+M phase.When combined use of VP andDHAD,the block effect of DHAD on cell cycle traverse was further enhanced and a striking increase of cell accumu-lation in the G_2+M phase was found.These results indicated that VP can pontentiate the DHAD—induced pertur-bation of cell cycle traverse and enhance cytotoxity of DHAD.
