血浆中甲硝唑前体水解产物HPLC测定法及水解动力学研究

Study on Hydrolytic Kinetics of Metronidazole-Phosphate in Rabbit Plasma by HPLC

本文建立了同时测定血浆中甲硝唑前体(甲硝唑磷酸酯)和母体的HPLC法,该法简便灵敏,重现性好。用此法研究了注射用甲硝唑前体制剂的水解动力学,在体外兔血浆中(37℃)甲硝唑前体制剂的水解过程为一级反应,水解速度常数K为0.016min^(-1),水解半衰期为43分钟。

A HPLC method for thesimultaneous determination of metroni-dazole and metronidazole-phosphate inrabbit plasma has been established. Thismethod is simple, sensitive, reproducibleand has a good linearity. It has been usedto study the hydrolytic kinetics of metro-nidazole-phosphate in rabbit plasma. Theresults show that hydrolytic process ofmetronidazole-phosphate in vitro at 37℃follows a first order kinetics with the rateconstant 0.016 min^(-1) and half-life 43 min.
