摘要
作者用醋酸冲洗膀胱粘膜,所得冲洗液经反相高压液相层析纯化出泛素样分子,N-端部分氨基酸序列测定显示其N-端25个氨基酸序列与人泛素一致。结合粘膜上皮抗菌多肽分子的发现,作者由此提出粘膜上皮也可能是一类免疫细胞的新见解。
A Ubiquitin-like peptidewas accidentally isolated from rat bladderby using 5% acetic acid wash while wewere isolating antibacterial peptides.Thepurified molecule was obtained by reversephase high performance liquid chromato-graphy. Gas phase microsequence analysisindicated the N-terminal sequences of themolecule as follows: MET-GLN-ILE-PHE-VAL-LYS-THR-LEU-THR-GLY-LYS-THR-ILE-THR-LEU-GLU-VAL-GLU-PRO-SER-ASP-THR-ILE-GLU-ASN. which is homo-logous to human ubiquitin. Ubiquitinplays a role in the differentiation of pre-B lymphocytes, Thus. it is suggestedfrom the findings of this molecule andthe endogenous antibacterial polypeptidesin mucosa or mucosal epithelium thatmucosal epithelium also might be one ofimmune cells or immunity-associated cells.which may secrete effector molecules dir-ectly to kill adherent microbes and pro-duce regulating factors in mucosal im-mune response.
