摘要
本文应用Disc-PAGE对豫南罗索线虫和食蚊罗索线虫寄生后期幼虫及成虫的4种酶的同工酶及应用SDS-PAGE对两虫种的蛋白区带进行比较分析,结果显示两虫种的LDH和MDH同工酶谱有明显差异,但也有相同位置的区带,过氧化物酶(PO)和脂酶(EST)没有区带呈现。两虫种的蛋白电泳谱较为接近,但在区带数目、主带分子量、次带的宽窄和染色深浅上均有不同,提示两虫种在遗传学上有一定联系,但又有显著差别。本研究表明同工酶和蛋白质电泳分析是罗索线虫分类鉴定和阐明虫种间亲缘关系的有效工具之一。
Isoenzyme electrophoreticpatterns of lactate dehydrogenase (LDH),malate dehydrogenase (MDH), esterase(EST) and peroxidase(PO)of postparasiticjuveniles and adults of Romanomermisyunanensis and R. culicivorax were ana-lysed by disc electrophoresis, respectively.The protein patterns of the two species ofRomanomermis were compared by verticalslab SDS polyacramide gel electrophoresis.The results showed marked differences inisoenzyme patterns(number of bands and/or Rf) of LDH and MDH between thetwo species of Romanomermis, but the simi-lar positions of isoenzyme bands denoteda certain genetic relationship betweenthem.No band was found in the isoenzymepatterns of PO and EST. Distinct dif-ferences in protein pattern, were observedbetween the two species of Romanomer-mis, but some common bands in patternreflected the phylogenetic relationships ofthese species.The differences of isoenzymeand protein patterns observed in thisstudy have provided the reason for iden-tifying and differentiating the two speciesof Romanomermis at the molecular level.
