本文报道α-1酸性糖蛋白的分离纯化。人血清经DEAE-Sephadex-A50和ReactiveBlue 2-Sepharose CL-6B两次柱层析即可获得纯化的α-1酸性糖蛋白。纯化产物经聚丙烯酰胺凝胶电泳、SDS-聚丙烯酰胺梯度凝胶电泳、免疫电泳和琼脂双向扩散等证明,其纯度和特异性均符合要求。

Alpha-1 acid glycoprotein,also named orosomucoid (ORM),is a serumprotein with genetic polymorphism. Thispaper describes a two-step procedure forpurifying ORM from human sera. The proce-dure consists of both anion exchange chro-matography on a DEAE-Sephadex-A50eolumn and affinity chromatography on aReactive Blue 2-Sepharose CL-6B column.The purified ORM identified by polyacry-lamide gel electrophoresis, SDS-polyacryla-mide gradient gel electrophoresis, immuno-electrophoresis and immunodiffusion washomogeneous and reacted specifically witha commercial anti-ORM serum (Sigma). Themolecular weight of the purified ORM wasabout 41 kd.
