摘要 目的 :探讨内皮细胞生长状态与血管平滑肌细胞增殖迁移能力的关系。方法 :通过建立细胞共培养体系 ,检测不同内皮生长状态对血管平滑肌细胞3 H TdR掺入、增殖细胞核抗原 (PCNA)蛋白表达、细胞迁移计数和α SM actinmRNA表达的影响。结果 :融合生长内皮使平滑肌细胞3 H TdR掺入量和PCNA蛋白表达明显降低 ,上调平滑肌细胞α SM actinmRNA表达 ;而对数生长内皮细胞使平滑肌细胞3 H TdR掺入量和PCNA蛋白表达明显升高 ,下调平滑肌细胞α SM actinmRNA表达。对照组平滑肌细胞在基础状态下存在少量迁移 ,对数增殖内皮细胞组平滑肌迁移数比对照组增高约 4倍 (P <0 .0 5 ) ,而融合生长内皮细胞组平滑肌迁移数仅为对照组的 0 .5倍 (P <0 .0 5 )。结论 :内皮细胞生长状态不同 ,对平滑肌细胞生物学特性的影响也不同 ,增殖期内皮明显促进平滑肌细胞增殖迁移、下调平滑肌细胞α SM actinmRNA表达。 AIM: To investigate the relation between the altered endothel ia l cells (EC) growth status and the proliferation and migration properties of vas cular smooth muscle cells(SMCs). METHODS: In the cell coculture sy stem, ECs were seeded on the lower chamber and SMCs on the upper chamber. 3H -TdR incoporation and PCNA protein expression were used to determine the effect s of varied EC growth states on the proliferation of vascular SMCs. The number of SMCs migration was counted. RT-PCR was used to analyze the influence of vari ed EC growth states on the expression of α-SM-actin mRNA. RESULTS: 3H-TdR incorportation and the expression of PCNA protein were signifi cantly lower in the confluent EC group and significantly higher in the prolifera tive EC group. The number of migrating cells was about 4 times higher in the proliferative EC group(n=6,P<0.05)than in the control group,while tha t in the confluent EC group was only half of the number of the control(n= 6,P<0.05). The proliferative ECs inhibited and confluent EC promoted signi ficantly the expression of α-SM-actin mRNA. CONCLUSION: These r esults suggest that the biologic features of SMCs are influenced by EC growth s tate. Proliferative ECs promoted significantly the proliferation and migration o f SMCs and down-regulate α-SM-actin mRNA expression.
