摘要
目的 :研究胆固醇 (CL)对大鼠心脏成纤维细胞增殖的影响 ,并对其机制进行探讨。方法 :分离、纯化、培养乳鼠心脏成纤维细胞 ,完全随机分为 3组 :对照组、中浓度胆固醇组 (0 .5g/L)和高浓度胆固醇组 (1.2g/L)。以3 H TdR掺入法测定各组细胞的增殖情况 ,并以流式细胞仪 (FCM)测定细胞周期。采用免疫组织化学染色方法观察各组c fos的表达情况。结果 :中浓度CL可使培养的乳鼠心脏成纤维细胞3 H TdR掺入量显著升高 (P <0 .0 5vs对照组 ) ,流式细胞仪检测表现为S期细胞明显增多。高浓度CL则抑制细胞生长 ,3 H TdR掺入量显著降低 (P <0 .0 5vs对照组 ) ,S期细胞数明显减少。同时中浓度CL显著升高Fos样免疫阳性细胞数及染色强度 (P <0 .0 5vs对照组 )。结论 :中等浓度胆固醇可促进大鼠心脏成纤维细胞增殖 ,其机制可能与影响细胞内c fos基因的表达有关。
AIM: To investigate the effect of cholesterol(CL) on the proliferation of the cardiac fibroblasts cultured from neonatal rats.MET HODS: The cultured rat cardiac fibroblasts were divided randomly into three groups:control group, middle concentration group (0.5 g/L CL) and high concentration group (1.2 g/L CL). The growth of rat cardiac fibroblasts in each group was measured by means of the incorporation of 3H-TdR and cell phases were determined by flow cytometer(FCM) after the fibroblasts were affected by water-soluble cholesterol. The effect of cholesterol on the intracellular level of c-fos was measured by means of immunohistochemistry stain.RES ULTS: The incorporation of 3H-TdR of rat cardiac fibroblasts of the mid dle concentration group(0.5 g/L) significantly increased (P<0.05 vs control group). It was indicated in FCM that the number of rat cardiac fibroblasts in S-phase increased significantly, however, CL of high concentratin(1.2 g/L) inhibited the proliferation of rat cardiac fibroblasts with the incorporation of 3H-TdR decreased(P<0.05 vs control group). CL of middle concentration( 0.5 g/L) significantly increased the expression of c-fos in rat cardiac fibroblasts(P<0.05, vs control group).CONCLUSION: CL of middle concentration can promote the proliferation of rat cardiac fibroblasts which is associated with the change in c-fos.
