摘要 目的 研究原发性肝癌及其它肿瘤组织中 gpc3/mxr7基因的表达与临床病理指标的关系。方法 收集正常成人肝、肾、肺等组织 ,原发性肝癌组织 79例和其它肿瘤组织 9例 ,采用Northern杂交方法 ,检测 gpc3/mxr7基因的表达 ,并分析其与临床病理指标的相关性。 结果 Northern杂交显示 gpc3/mxr7基因在正常肝脏组织中不表达 ,在肾、肺中有低丰度的表达。 79例肝癌组织中 gpc3/mxr7基因阳性率为 73 4 1% (5 8/ 79) ,明显高于其癌旁组织 (6 32 % ,5 / 79,P <0 0 1) ,其阳性率与肝癌的大小和恶性程度呈正相关 (P <0 0 5 )。gpc3/mxr7基因在其它肿瘤中均不表达。结论 gpc3/mxr7基因可能是具有肝癌组织特异性的高表达基因 ,可望成为临床肝癌诊断、治疗和预后的辅助参考指标。 Objective To investigate the relationship between clinical features and expression of gpc3/mxr7 gene in primary hepatocellular carcinoma (HCC) and other tumors. Methods Recombinant plasmid pBSKII-gpc3 was constructed by inserting the gpc3/mxr7 coding sequences into the clone vector pBSKII. Using 32P-labeled gpc3/mxr7cDNA as the probe, gene expression was detected by Northern blot analysis. Results gpc3/mxr7 mRNA was undetectable in liver and was expressed in low level in lung and kidney in adults. However, the detectable rate of gpc3/mxr7 mRNA was 73.41% in the 79 cases of HCC, which was higher than in pericancerous tissues. Meanwhile, the positive rate was positively correlated to the tumor size and malignancy. The expression of gpc3/mxr7 mRNA was not detected in other tumors. Conclusions gpc3/mxr7 might be a kind of specific gene in HCC and can be used as a maker for diagnosis, treatment and prognosis of HCC.
