摘要
目的 观察生长抑素对大鼠移植性肝癌血管内皮生长因子 (VEGF)、基质金属蛋白酶 1(MMP 1)表达的影响 ,初步探讨生长抑素抑制大鼠移植性肝癌生长的机制。方法 制作大鼠移植性肝癌的动物模型 ,随机分为 2组 ,对照组 (n =11) ,治疗组 (n =8)。治疗组腹腔注射善宁 (10 0 μg·kg-1·d-1) ,每天分 2次注射 ,对照组注射等量生理盐水。 10d后心脏采血 0 5ml后处死大鼠 ,大鼠血液室温下 4h ,待其凝固 ,离心 (30 0 0转 /min) 10min ,分离血清放入 - 80℃冰箱保存。酶联免疫吸附试验检测血清VEGF ,原位杂交法检测瘤组织VEGF、MMP表达。结果 治疗组血清VEGF值低于对照组 ,瘤组织VEGFmRNA、MMP 1mRNA在对照组、治疗组中均有阳性表达 ,而治疗组表达水平较对照组低 (P <0 0 5 )。结论 生长抑素抑制血清VEGF ,转移相关基因VEGF、MMP 1表达降低 ,可能是生长抑素抑制大鼠移植性肝癌生长的机制之一。
Objective To explore the effects of somatostatin on expression of MMP and VEGF in rats with transplanted liver cancer (TLC) and to evaluate its anti-angiogenesis effect. Methods After the model of TLC was established in 19 rats, the animals were randomized into the treated group (n=8) and the control group (n=11). The rats in the treated group received subcutaneous injection of somatostatin (100 μg·kg^(-1)·d^(-1), bid) and those in the control group received the same amount of normal saline. All rats were put to death 10 days later after collection of 0.5 ml blood in each. The serum samples were separated and preserved under -80℃. ELISA was applied to detect VEGF level. The expression of VEGF and MMP in tumor tissue was determined by in situ hybridization. Results The serum VEGF level and expression of VEGF and MMP in tumor tissue were significantly lower in the treated group than in the control (P<0.05). Conclusions Somatostatin can decrease the serum VEGF level and may inhibit angiogenesis. Somatostatin may also inhibit expression of VEGF and MMP-1 that are metastatic related genes.
