摘要
目的 明确三氧化二砷对人肝癌细胞体外生长的影响 ,并探讨其作用机制。方法 采用四甲基偶氮唑蓝法、3 H TdR掺入法、TUNEL法、透射电镜观察及流式细胞术 ,观察不同浓度的三氧化二砷对体外培养的人肝癌细胞株SMMC 772 1的生长活力、细胞凋亡、DNA合成能力的影响。结果 三氧化二砷能显著地抑制人肝癌细胞SMMC 772 1的生长 ,并且具有时间和剂量依赖性 ;发生作用后 ,细胞生长有明显的凋亡特征性改变 ;细胞DNA合成能力下降。结论 三氧化二砷能有效地抑制体外肝癌细胞生长 。
Objective To investigate the effects of arsenic trioxide on proliferation and apoptosis of human hepatocellular carcinoma (HCC) line and explore its molecular mechanism. Methods The effects of arsenic trioxide on proliferation, apoptosis and DNA synthesis of SMMC-7721 cells were determined by MTT assay, ~3H-TdR incorporation into DNA, TUNEL, electron microscopy and flow cytometry. Results The growth of SMMC-7721 cells was inhibited by As 2O 3 in a time/dose-dependent manner. As 2O 3 induced apoptosis of SMMC-7721 cells and reduced their DNA synthesis. Conclusions As 2O 3 is effective in inhibiting growth of HCC in vitro. The mechanism of antineoplastic effect of As 2O 3 might be inhibition of DNA synthesis and induction of apoptosis of tumor cells.
