摘要
目的:检测人白细胞介素-23(IL-23)和白细胞介素-12(IL-12)亚基在各类白血病细胞系中的表达并探讨佛波二酯对IL-23和IL-12亚基表达的调控作用。方法:半定量RT-PCR方法检测13株人白血病细胞系IL-23p19,IL-12p35,IL-12p40 亚基的表达。用佛波二酯处理白血病细胞系,不同时间点取样,半定量RT-PCR方法检测上述亚基的表达变化。结果:检测的大多数白血病细胞系均表达p19 mRNA,p19比p35表达更为普遍;而p40仅在EB病毒感染的LCL和J6-1细胞中测到。佛波二酯可以提高EB病毒阳性细胞系中p19,p35和p40的表达水平。结论:p19可能广泛表达于各类血细胞。佛波二酯可上调EB病毒阳性细胞系p19,p35和p40的表达,表明IL-23与IL-12相类似,其在白血病细胞系中的表达可能与EB病毒感染相关。
Objective: To detect expression of interleukin- 23 (IL- 23) and IL- 12 subunits in various leukemia cell lines and evaluate the ability of phorbol diester to up- regulate the subunits expression. Methods: Expression of IL- 23p19, IL- 12p35 and IL- 12p40 in 13 leukemia cell lines both constitutively and upon stimulation with phorbol diester was studied by semiquantitative RT-PCR. Results: p19 mRNA was detectable in most leukemia cell lines tested but p35 distribution was more restricted. p40 was expressed only by Epstein-Barr virus-positive (EBV+) cell lines LCL and J6-1. Phorbol diester enhanced p19, p35 and p40 expression in EBV+ cell lines. Conclusion: IL- 23 p19 expression was ubiquitously expressed in blood cells. All the three subunits of IL- 23 and IL- 12 were strongly inducible by phorbol diester in EBV+ cell lines, suggesting that, as IL- 12, expression of IL- 23 subunit in leukemia cell lines also depend on EBV infection.
