免疫复合物对U937细胞和中性粒细胞表面Fc受体表达的调节

摘要 目的探讨免疫复合物(ICs)对U937细胞和中性粒细胞表面Fc受体表达的调节。方法将U937细胞或中性粒细胞与各种免疫复合物共同孵育1h后,用流式细胞仪分析细胞表面各种Fc受体的表达情况。结果IgG1ICs和IgG3ICs能上调U937细胞表面FcγRⅡ和FcγRⅢ以及中性粒细胞表面的FcγRⅠ和FcαRⅠ的表达。与各类免疫复合物孵育后中性粒细胞表面FcγRⅡ的表达呈降低趋势。结论免疫复合物能调节中性粒细胞和U937细胞Fc受体的表达,其中以IgG1ICs和IgG3ICs最为明显。 Objective To study the regulation of Fc receptor expression by immune complexes?(ICs?)on neutro-phils and U937 cells. Methods IgA ICs? IgG1 ICs? IgG2 ICs? IgG3 ICs? IgG4 ICs? and IgM ICs were incubated with neutrophils or U937 cells for 1 h. Then their surface Fc receptors were stained by anti-FcγRⅠ? anti-FcγRⅡ? anti-FcγRⅢ? and anti-FcαRⅠmonoclonal antibodies and analyzed by fluorescent activated cell sorting?(FACS?). Results IgG1 ICs and IgG3 ICs up-regulated FcγRⅡ and FcγRⅢ on U937 cells? FcγRⅠand FcαRⅠ on neutrophils. Almost all ICs down-regulated FcγRⅡ on neutrophils. Conclusions ICs can regulate Fc receptor expression on neutrophils and U937 cells? among which IgG1 ICs and IgG3 ICs are most effective.
